  三、主讲内容

  (一))主题一:外泌体在HIV-1和吗啡滥用诱导的脑炎中的调控作用

  主讲人:美国内布拉斯加大学医学中心教授 Shilpa J.Buch

  内容简介:

  The objective of the current study was to examine the roles of miRs released from EVs isolated from HIV Tat and morphine exposed astrocytes, in mediating synaptodendritic injury and microglial activation, respectively. Our data demonstrated that exposure of astrocytes to HIV Tat induced the expression and secretion of miR-9 in the EVs, which, upon uptake by the neurons, resulted in impaired spine density when assayed by confocal microscopy. Additionally, miR-7/-138 were induced in the EVs released by morphine exposed astrocytes, which, upon uptake by microglia, led to their activation, via binding and activation of the Toll-like receptor 7/ NF-kB nuclear translocation pathway. In conclusion our findings indicate that HIV Tat and morphine mediated dysregulation of miRNA expression in the brain involves cell-cell communication via the extracellular vesicles, which in turn, leads to synaptodendritic injury and microglial activation.

  主讲人简介:

  Dr. Shilpa Buch is a professor and vice chair in the department of pharmacology and experimental neuroscience at the University of Nebraska Medical Center. She is also the director of the Nebraska Center for Substance Abuse Research at UNMC. Her primary research focus is aimed at understanding the molecular mechanism(s) by which drugs of abuse, such as cocaine and morphine, co-operate with HIV-1/HIV proteins to enhance progression of HIV-1 associated neurological disorders (HAND). More recently, her research is centered on exploring the molecular pathways by which microRNAs, small noncoding RNAs that shuttle in extracellular vesicle cargos, regulate gene expression and cellular cross-talk.

Over the past 15 years, Dr. Buch has been an active member of NIH study sections and considers it her privilege to “finally be able to give back to NIH after continuing to be its beneficiary for many years.” Dr. Buch has won numerous awards for her research and has published extensively in the field of HIV and drug abuse.

  主讲人:暨南大学生物学组团一级特聘研究员 王通

  主讲人简介:王通,博士,研究员,博士研究生导师,暨南大学生物学组团一级特聘研究员。中国蛋白质组专业委员会(CNHUP0)理事, CNHUP0第一届青年委员会主任委员,广东省外科学重点学科肿瘤基础与临床方向学术帯头人,美国化学学会(ACS)会员奖,美国UW/FHCRCCFAR国际会员和新研究学者奖获得者,广东省企业科技特派员。截至2017年6月,以通讯作者或第一作者在Nucleic Acids Research、PLoS Genetics和 Journal of Proteome Research等国际期刊发表论文21篇,主持包括十三五国家科技重大专项,国家自然科学基金,广东省应用型科技研发重大专项和广州市产学研协同创新重大专项等国内外重大课题20项。应邀在包括美国UW/FHCRC CFAR年会、韩国人类蛋白质组组织年会、新加坡下一代测序年会、香港国际癌症年会(HKICC) 和中国计算蛋白质组学研讨会等多个国际会议上做大会口头报告。是GastroenteroIogy, J. Proteome Res., Sci Reports, Proteomics, BMC Genomics, Mol. BioSyst., J. Proteomics和 PloS One等1 0余种国际期刊的审稿人 。
